Personal Hygiene in the Cell Culture Laboratory

Maintaining proper personal hygiene is essential when working in a cell culture laboratory.

Poor hygiene practices can introduce contamination during the manipulation of cells, media, and reagents.

Hands

Hand hygiene is one of the most important factors in preventing contamination.

Handwashing with soap and water is effective in reducing the skin microbiota, which represents one of the main sources of contamination in cell culture (see i.e.here or here).

Sterile technique must always be supported by proper hand care.

Gloves

Gloves should always be worn when handling cells and reagents to protect both the user and the samples.

However, it is important to remember that gloves are not sterile by default and they can easily become contaminated during use

For this reason disinfect gloves frequently using 70% ethanol, change gloves when moving outside the laminar flow hood, change them again before returning to sterile work

Gloves should also fully cover the wrists. A good practice is to position the lab coat sleeves under the glove cuffs to ensure complete coverage.

 

Hair

Long hair should be tied back to prevent accidental contamination of sterile areas.

Face and behavior

When working under a laminar flow hood avoid talking as much as possible, if necessary, speak away from the opening of the hood, use the front glass as a barrier

To reduce contamination risks: do not enter the laboratory if you are ill, avoid coughing or sneezing near sterile work areas, wearing a face mask can provide additional protection

Lab coat

The lab coat used during cell culture procedures must be clean and dedicated exclusively to the cell culture laboratory. It should not be worn outside the lab to prevent cross-contamination.

Additional considerations

In more controlled environments (e.g. GMP laboratories), stricter protective measures may be required, including masks, hair covers or additional protective equipment

In standard research laboratories, these measures may not always be necessary, but good hygiene practices remain essential.

Conclusion

Good personal hygiene is a fundamental part of aseptic technique.

Even small lapses in hygiene can compromise sterility and affect experimental results.

Maintaining consistent and careful habits is key to successful cell culture work.
